Beyond its established role in puberty and fertility, recent human trials at Imperial College London found kisspeptin administration increased activity in brain regions governing sexual desire and arousal in both men and women with low libido — with no reported side effects, a notable finding for early-stage drug development. - Melanotan I —FDA-approved since 2019 as Scenesse (afamelanotide), for erythropoietic protoporphyria. Selectively activates MC1R, driving melanin production (makes you tan)with a well-documented Phase 3 evidence base (median 69.4 pain-free sun-exposure hours vs. 40.8 on placebo).
- Melanotan II — A non-selective melanocortin receptor agonist, activating MC1R, MC3R, and MC4R. Originally developed as a tanning agent, but early trials found it also triggered spontaneous sexual arousal via its MC3R/MC4R activity — an observation that directly led to the development of PT-141 as a more selective analog. No approved indication anywhere; documented case reports include melanoma association, priapism, rhabdomyolysis, and renal infarction.
- PT-141 (Bremelanotide) —FDA-approved since 2019 as Vyleesi, for hypoactive sexual desire disorder. A more MC4R-selective analog of Melanotan II, developed specifically to retain the arousal effect while reducing the broader pigmentation/off-target activity.
